The determination of serum acid and alkaline phosphatase activity with 4-aminoantipyrine (A.A.P.).

چکیده
C6H5 Gottlieb and Marsh (1946), using alka:ine ferricyanide as the oxidant, devised a method for the estimation of certain phenolic fungicides with A.A.P. The procedure was modified by Grifols (1951) for the determination of phenol in the measurement of alkaline phosphatase activity in body fluids. This author made a careful study of the factors which control the development of the colour and his results showed good correlation with those obtained by the method of King (1951). Advantages claimed for the method were that the reagent did not react with proteins, thus obviating their precipitation during the estimation, and also the colour development was comparatively rapid. The use of A.A.P. as a reagent for phenol appeared to be of potential value especially if its use could be extended to the estimation of serum acid phosphatase activity. Considerable modification of the original Grifols procedure was found to be necessary in order to measure both alkaline and acid phosphatase activity in blood. The use of calibration curves, the stability of the colour, and the effect of the presence of serum

Automated determination of acid phosphatase. II. Technic for using robot chemist.
A procedure is presented for the automated determination of acid phosphatase activity in biological materials using the Robot Chemist. Although either phenylphosphate and a-naphthylphosphate may be used as substrate in this analysis, the procedure is described in detail for serum acid phosphatase using a-naphthylphosphate in 0.1 M citrate, pH 5.2, since this substrate is more selective for pros...

A procedureis presentedfor the automated determination of acid phosphatase activity in biological materials from either plant or animal origin. The Technicon N-i flow diagram has been simplified, and the reagents for the measurement of enzymatically produced phenolic substanceshave been modified without lossof range or sensitivity. Both phenyiphosphateand a-naphthylphosphate, introduced by Babs...
متن کاملP-178: Separation and Identification of Alkaline Phosphatase Isozymes during Pregnancy
Background: Alkaline phosphatase (ALP), (EC 3.1.3.1) is a hydrolase enzyme responsible for removing phosphate groups from various molecules in the body. In humans, ALPis present in all tissues such as liver, bile duct, kidney, bone, and the placenta which detection of its activity is so useful in molecular biology. Pregnancy is associated with normal physiological changes that assist fetal surv...
متن کاملA method for the determination of tartratelabile, prostatic acid phosphatase in serum.
L(+)-Tartrate strongly inhibits prostatic acid phosphatase (Abul-Fadl and King, 1948, 1949) while having no action on red cell phosphatase. The authors suggested the application of this phenomenon and its diagnostic value as a sensitive test in elucidating doubtful or borderline cases of prostatic cancer. Based on this principle, Fishman and Lerner (1953) described a method for the determinatio...
متن کاملThe effects of chromium and cadmium on the activity of alkaline phosphatase lsoenzmes in rats
The aim of this study was to Investigate the toxic effects of chromium and cadmium on the activity of two isoenzymes of alkaline phosphatase in vitro and in vivo experiments. Oral administration and peritoneal injection of CdCl2 or CrCl3 in rats reduced serum alkaline phosphatase by 21.5℅ and 25.5℅ respectively. At the same time high molecular weight alkaline phosphatase was elevated by 138℅ a...
